ABSTRACT

Background: Ayanin (3,7,4'-Tri-O-methylquercetin) and 3,7-Di-O-methylquercetin (DMQ) are the main active metabolites isolated by bioguided fractionation from Croton schiedeanus, species known popularly in Colombia as "almizclillo", which has been studied in experimental models in rats, exerting vasodilator and antihypertensive effects. Also, when the effect of these flavonoids was studied separately, important vasodilation was observed. Objective:To evaluate whether flavonoids from Croton schiedeanus have synergistic vasodilator properties when different combinations are used in isolated aorta rings. Methods: Cumulative concentrations of ayanin (10-8 M - 6x10-5 M or 0.01 µM - 60 µM) were assayed in the absence and presence of an increasing concentration of 3,7-Di-O-methylquercetin (DMQ) (10-8 ­ 3x10-5M or 0.01­30 µM) in isolated rings from Wistar rats, pre-contracted with phenylephrine. The concentration-response curve with the maximal effect was compared with that obtained by Croton schiedeanus whole ethanolic extract (10-6 ­ 3x10-4 g/mL). Also, this combination was assayed in the presence of the nitric oxide synthetase inhibitor L-NAME (10-4 M) and the guanylate cyclase inhibitor methylene blue (10-4 M) to assess the role of the NO/cGMP pathway in this interaction. Results: Ayanin and DMQ display a dual interaction in vascular relaxant response: agonism at higher concentration ranges (10-6 ­ 3x10-5 M or 1­30 µM) and antagonism at lower concentration ranges (10-8 ­ 3x10-7 M or 0.01­0.3 µM). The efficacy at the highest concentration was greater than that obtained by the whole extract (Emax: 98.4% vs. 33.9%). This response was decreased but not reverted in the presence of L-NAME and methylene blue. Thus, the vasodilator effect of this combination does not depend entirely on the NO/cGMP cyclic pathway. Conclusion: The combined use of appropriate concentrations of these flavonoids could represent an advantage over Croton schiedeanus whole extract for vasodilator purposes


Antecedentes: Ayanina (3,7,4'-Tri-O-metilquercetina) y 3,7-Di-O-metilquercetina (DMQ) son los principales metabolitos activos aislados mediante fraccionamiento bioguiado, a partir de Croton schiedeanus, especie conocida popularmente en Colombia como "almizclillo", la cual ha sido estudiada en modelos experimentales en ratas, ejerciendo efectos antihipertensivos y vasodilatadores. Además, al estudiar por separado el efecto de los flavonoides, se observó importante vasodilatación. Objetivo:Evaluar si los principales flavonoides de Croton schiedeanus tienen propiedades vasodilatadoras sinérgicas al utilizar diferentes combinaciones de ellos en anillos de aorta aislados. Metodología: Se analizaron concentraciones acumulativas de ayanina (10-8 M - 6x10-5 M o 0,01 µM - 60 µM) en ausencia y en presencia de concentraciones crecientes de DMQ (10-8 M - 3x10-5 M o 0,01 µM ­ 30 µM) en anillos aislados de ratas Wistar, pre-contraídos con fenilefrina. La curva concentración respuesta obtenida con el efecto máximo, fue comparada con la obtenida con el extracto etanólico de Croton schiedeanus (10-6 - 3x10-4 g/mL). Adicionalmente, esta combinación fue ensayada en presencia del inhibidor de óxido nítrico sintetasa L-NAME (10-4 M) y el inhibidor de guanilato ciclasa, azul de metileno (10-4 M) para evaluar el papel de la vía NO/GMPc en esta interacción. Resultados: Ayanina y DMQ muestran una interacción dual en la respuesta vascular relajante: agonismo en el rango más alto (10-6 M ­ 3x10-5 M o 1 µM ­ 30 µM), y antagonismo en el rango más bajo (10-8 M ­ 3x10-7 M o 0.01 µM ­ 0,3 µM). A altas concentraciones, la eficacia de los flavonoides fue mayor que las obtenidas por el extracto completo (Emáx: 98,4% vs 33,9%). Esta respuesta disminuyó, pero no se revirtió en presencia de L-NAME y azul de metileno. Por lo tanto, el efecto vasodilatador de esta combinación no depende enteramente de la vía NO/GMPc. Conclusión: El uso combinado de las concentraciones apropiadas de estos flavonoides podría representar una ventaja sobre el extracto de Croton schiedeanus, con propósitos vasodilatadores

ABSTRACT: Pea (Pisum sativum L.) is the fourth leading legume crop in the world, and its demand is increasing. In this study, the morphological characteristics (seed shape, seed surface, seed coat color, hilum color, cotyledon color, 100-seed weight and color values), total phenolic content (TPC), total flavonoid content (TFC), 2,2'-azinobis-(3-ethylbenzthiazoline-6-sulphonate) (ABTS) free radical scavenging capacity and ferric reducing antioxidant power (FRAP) of 75 pea cultivars were investigated. Results showed rich genetic diversity and a wide range of phenolic contents and antioxidant activities. Sixteen varieties enriched with phenolic contents and high antioxidant activities were screened out. A significant correlation was reported among color values, TPC, TFC, ABTS and FRAP. Principal component analysis (PCA) extracted four principal components with a total cumulative contribution of 81.29%. Hierarchical cluster analysis based on the four extracted principal components resulted in a dendrogram dividing the peas into three groups. In addition, dark pea seeds have potential as a functional food in addition to their traditional role in providing dietary protein and fibre. This study provided a scientific basis for the breeding of pea varieties, development of new products and improvement of pea resource utilization.


RESUMO: A ervilha (Pisum sativum L.) é a quarta cultura de leguminosas líder no mundo e sua demanda está aumentando. Neste estudo, as características morfológicas (forma da semente, superfície da semente, cor da casca, cor do hilo, cor do cotilédone, peso de 100 sementes e valores de cor), teor fenólico total (TPC), teor de flavonóides totais (TFC), 2,2. A capacidade de eliminação de radicais livres de '-azinobis- (3-etilbenzotiazolino-6-sulfonato) (ABTS) e o poder antioxidante redutor de ferro (FRAP) de 75 cultivares de ervilha foram investigados. Os resultados mostraram rica diversidade genética e uma ampla gama de conteúdos fenólicos e atividades antioxidantes. Dezesseis variedades enriquecidas com conteúdo fenólico e alta atividade antioxidante foram descartadas. Uma correlação significativa foi encontrada entre os valores de cores, TPC, TFC, ABTS e FRAP. A análise de componentes principais (PCA) extraiu quatro componentes principais com uma contribuição total acumulada de 81, 29%. A análise hierárquica de agrupamento foi baseada nos quatro componentes principais extraídos resultou em um dendrograma dividindo as ervilhas em três grupos. Assim, as sementes de ervilha escura têm potencial como alimento funcional, além de seu papel tradicional no fornecimento de proteína e fibra dietética. Este estudo fornece uma base científica para a criação de variedades de ervilha, desenvolvimento de novos produtos e melhoria da utilização de recursos de ervilha.

ABSTRACT

Ultraviolet radiation (UV) is related to the development of skin cancer and photoaging, also produces free radicals or reactive oxygen species (ROS) that cause premature aging. Therefore, sunscreens with extended UV protection capacity have been studied associated with antioxidant compounds. Different components can be incorporated with the purpose of improving, the photoprotection efficiency, as well as other cosmetic attributes, creating a multifunctional product. Among the compounds, there are some to be used from natural origin that are excellent, because it reduces the side effects and toxicity. Flavonoids are good examples of natural agents, because have demonstrated photoprotective antioxidant action in food and it is also promising for topical use. The aim of this work is to develop photoprotective formulations with quercetin, evaluating potential photoprotective and antioxidant of each formulation through tests already established and described in the literature, with emphasis on in vitro testing of potential photoprotective, antioxidant and security (het-cam); ex vivo antioxidant potential (tape-stripping). The results show us the photoprotective and antioxidant capacity of quercetin and the ideal concentration for this potential, thereby contributing to the synthesis of new molecules and development of new products, concerning stable, safe and effective exposure to solar UV radiation, preventing new incidences of skin cancer and reduction of photoaging


A radiação ultravioleta (UV) está relacionada ao desenvolvimento de câncer de pele e fotoenvelhecimento, também produz radicais livres ou espécies reativas de oxigênio (EROs) que causam o envelhecimento precoce. Portanto, protetores solares com capacidade de proteção UV amplo espectro foram estudados associados a compostos antioxidantes. Diferentes componentes podem ser incorporados com o objetivo de melhorar a eficiência da fotoproteção, assim como outros atributos cosméticos criando um produto multifuncional. Entre os compostos a serem utilizados, de origem natural são excelentes, pois reduzem os efeitos colaterais e a toxicidade. Os flavonóides são bons exemplos de agentes naturais, porque demonstraram ação antioxidante fotoprotetora nos alimentos e também são promissores para uso tópico. O objetivo deste trabalho é desenvolver formulações fotoprotetoras com quercetina, avaliando potenciais fotoprotetores e antioxidantes de cada formulação através de testes já estabelecidos e descritos na literatura com ênfase em ensaios in vitro de potenciais fotoprotetores, antioxidantes e de segurança (het-cam); potencial antioxidante ex vivo (tape-stripping). Os resultados nos mostram a capacidade fotoprotetora e antioxidante da quercetina e a concentração ideal para este potencial, contribuindo para a síntese de novas moléculas e desenvolvimento de novos produtos estáveis, seguros e eficazes em relação à exposição à radiação solar UV, prevenindo novas incidências de câncer de pele e redução do fotoenvelhecimento

RESUMEN Objetivos . Evaluar el efecto antiinflamatorio de la fracción flavonoide de Lepechinia meyenii (Walp.) Epling sobre leucocitos de pacientes con artritis reumatoide (AR). Materiales y métodos. Se recolectaron plantas de la especie Lepechinia meyenii (Walp.) Epling extrayendo diferentes fracciones flavonoides por cromatografía de columna y de capa fina. Se evaluó la producción de anión superóxido mediante la técnica de ensayo reducción nitroblue tetrazolium, en neutrófilos obtenidos de sangre de pacientes con AR, separados en tres grupos: control negativo, que consistió de neutrófilos (5x105 células), control positivo, formado por neutrófilos activados con PMA (phorbol myristate acetate) (150 ng/mL) y los tratamientos, formados por neutrófilos activados y tratados con diferentes concentraciones de la fracción flavonoide LM8 (60, 120 y 180 ug/mL). La expresión de genes proinflamatorios se estudió por RTqPCR, en leucocitos mononucleares obtenidos de pacientes con AR separados en tres grupos: control negativo, que consistió de leucocitos mononucleares (5x105 células), control positivo formado por leucocitos mononucleares activados con fitohemaglutinina (PHA) (150 ug/mL) y el tratamiento formado por leucocitos mononucleares activados y tratados con la fracción flavonoide LM8 (120 ug/mL). Resultados . Se purificaron varias fracciones flavonoides, resultando la fracción LM8 con el mejor efecto inmunomodulador. Dicha fracción disminuyó la producción de anión superóxido en una manera dependiente de la concentración. Por otro lado, disminuyó la expresión de TNFα, IL8 e IL17 en leucocitos mononucleares. Conclusiones. Estos resultados son alentadores respecto al efecto inmunomodulador de esta planta medicinal peruana y justifican continuar su estudio para una posible aplicación clínica.


ABSTRACT Objectives. to assess the anti-inflammatory effect of the flavonoid fraction of Lepechinia meyenii (Walp.) Epling on leukocytes of patients with rheumatoid arthritis (RA). Materials and Methods. Plants of the species Lepechinia meyenii (Walp.) Epling were collected and then different flavonoid fractions were extracted by column and thin layer chromatography. The superoxide anion production was evaluated by means of the reduction of nitroblue tetrazolium assay technique in neutrophils obtained from the blood of patients with RA, divided into three groups: negative control, which consisted of neutrophils (5x105 cells); positive control, made up of PMA (phorbol myristate acetate)-activated neutrophils (150 ng/mL), and the treatments, comprised of neutrophils activated and treated with different concentrations of the flavonoid fraction LM8 (60, 120, and 180 ug/mL). The expression of pro- inflammatory genes was studied by RTqPCR in mononuclear leukocytes obtained from patients with RA, divided into three groups: negative control, which consisted of mononuclear leukocytes (5x105 cells); positive control, made up of phytohaemagglutinin (PHA) (150 ug/ml)-activated mononuclear leukocytes, and the treatment, comprised of mononuclear leukocytes activated and treated with the flavonoid fraction LM8 (120 ug/mL). Results. Several flavonoid fractions were purified, with fraction LM8 showing the best immunomodulating effect. Said fraction diminished the superoxide anion production dependent on concentration. On the other hand, it diminished the expression of TNFα, IL8, and IL17 in mononuclear leukocytes. Conclusions. These results are encouraging in terms of the immunomodulating effect of this Peruvian medicinal plant and justify the continuation of their study for a potential clinical application.

ABSTRACT

A exposição crônica à radiação solar pode contribuir para o aparecimento do câncer de pele, sendo o uso de fotoprotetores um fator primordial na prevenção desses efeitos deletérios. Atualmente, substâncias bioativas tais como a rutina têm sido foco de interesse da comunidade científica graças às suas propriedades fotoprotetoras e antioxidantes, que podem promover aumento dos valores de FPS, além de conferir características multifuncionais às formulações. Achados in vitro recentes indicam que a rutina, quando incorporada em emulsões fotoprotetoras óleo em água, promove aumento da atividade antioxidante e aumento do FPS. No entanto, a realização de estudos clínicos é fundamental para confirmar e quantificar esses resultados, já que a metodologia in vitro possui baixa repetibilidade e ausência de correlação com ensaios in vivo, principalmente quando as formulações analisadas apresentam substâncias antioxidantes em sua composição. O objetivo deste estudo foi avaliar pela primeira vez a atividade da rutina frente ao FPS e sua segurança clínica através da comparação de formulações fotoprotetoras contendo rutina 0.1% (w/w), avobenzona 3.0% (w/w) e octil dimetil PABA 8.0% (w/w) com uma preparação similar sem o composto bioativo. Adicionalmente, hidratação cutânea, FPS in vitro e atividade antioxidante da rutina em associação com outros filtros foram investigados. O perfil de segurança das formulações qualificou as fórmulas para os testes de eficácia clínica. O teste de DPPH confirmou a capacidade antioxidante da rutina, demonstrando cerca de 40% de aumento na capacidade de sequestro de radicais livres na presença do composto bioativo. A rutina em combinação com os filtros UV aumentou o FPS clínico de 7.30 ± 0.60 para 12.37 ± 1.13, o que representa cerca de 70% de aumento. Os resultados encontrados provam que a rutina em combinação com outros filtros pode aumentar significativamente o valor do FPS e que a mesma é segura para uso clínico


Unprotected chronic exposure to solar radiation can contribute to premature skin cancer and sunscreens are a key factor to avoid those detrimental effects. Currently, there is a growing interest in the photoprotector and antioxidant potential of bioactive substances, such as rutin, that could help to increase the SPF value and add multifunctional characteristics to the formulations. Recent in vitro findings indicated that rutin, when incorporated in oil-in-water photoprotective emulsions can provide antioxidant activity and SPF increase. However, clinical studies are fundamental to determine this activity duo to in vitro methodology lack of repeatability and correlation between the in vivo data, especially when the analyzed formulas contain antioxidant substances. The aim of this study was to evaluate for the first time to date the rutin in vivo SPF and clinical safety by comparing sunscreens formulations containing rutin 0.1% (w/w), butyl methoxydibenzoylmethane 3.0% (w/w) and octyl dimethylPABA 8.0% (w/w) with a similar bioactive-free preparation. Additionally, skin hydration, in vitro SPF and in vitro antioxidant activity of rutin, in association with the UV filters were investigated. The safety profile of the formulations under sun-exposed skin conditions qualified the formulas for clinical efficacy assays. DPPH test confirmed rutin antioxidant properties, demonstrating about 40% increase in radical scavenging potential when the bioactive compound was present. Rutin in combination with the UV filters increased the clinical SPF from 7.30 ± 0.60 to 12.37 ± 1.13, representing about 70% growth in the SPF value. The results obtained proved that rutin in combination with UV filters can improve the SPF value significantly and is safe for clinical use

The aim of this study was to evaluate the effect of different concentrations of trans-resveratrol or quercetin on the ability of goat sperm to withstand being frozen. Six pools of semen obtained from six male goats were processed with different concentrations of resveratrol or quercetin (Experiment 1: 0, 15, 25, 50, 75 or 100µM resveratrol; Experiment 2: 0, 15, 25, 50, 75 or 100µM quercetin) and frozen. After thawing, the semen was evaluated for sperm kinematics, plasma membrane and acrosome integrity, morphology and oxidative stress following 0 and 1h of incubation. Immediately after thawing (0h), wobble (oscillation index) in the groups treated with 100µM of quercetin or resveratrol was lower (P<0.05) than in those treated with 0 and 25µM resveratrol and 0µM quercetin, respectively. After 1h of incubation, the total motility in treatments with 15, 50 and 75µM quercetin, as well as the plasma membrane integrity in all quercetin concentrations were lower (P<0.05) than at 0h. In opposition, the linearity of semen samples treated with 100µM quercetin and the straightness of those treated with 75 and 100µM quercetin were lower (P<0.05) at 0h than at 1h after thawing. Thus, it can be concluded that resveratrol and quercetin at high concentrations (100µM) transiently reduce the wobble of goat sperm submitted to frozen storage, and that quercetin (75 and 100µM) increases the linearity and straightness over time, which can be favorable for fertility.(AU)


O objetivo deste estudo foi avaliar o efeito de diferentes concentrações de transresveratrol ou quercetina sobre a capacidade dos espermatozoides caprinos de resistirem à congelação. Seis pools de sêmen, obtidos de seis reprodutores caprinos, foram processados com diferentes concentrações de resveratrol ou quercetina (Experimento 1: 0, 15, 25, 50, 75 ou 100µM de resveratrol; Experimento 2: 0, 15, 25, 50, 75 ou 100µM de quercetina) e congelados. Após o descongelamento, o sêmen foi avaliado quanto à cinética espermática, à integridade das membranas plasmática e acrossomal, à morfologia e ao estresse oxidativo nos tempos zero e uma hora de incubação. Imediatamente após a descongelação (zero hora), o wobble (índice de oscilação) nos grupos tratados com 100µM de quercetina ou de resveratrol foi menor (P<0,05) do que nos tratados com 0 e 25µM de resveratrol e com 0µM de quercetina, respectivamente. Após uma hora de incubação, a motilidade total dos tratamentos com 15, 50 e 75µM de quercetina, assim como a integridade de membrana plasmática em todas as concentrações de quercetina, foi menor (P<0,05) do que à zero hora. Em oposição, a linearidade das amostras de sêmen tratadas com 100µM de quercetina e a retilinearidade daquelas tratadas com 75µM e 100µM de quercetina foram menores (P<0,05) à zero hora do que à uma hora após descongelação. Assim, pode-se concluir que o resveratrol e a quercetina, em concentrações elevadas (100µM), reduzem, transitoriamente, o índice de oscilação de espermatozoides caprinos submetidos à congelação e que a quercetina (75 e 100µM) aumenta a linearidade e a retilinearidade ao longo do tempo, o que pode ser favorável à fertilidade.(AU)

ABSTRACT

INTRODUCCIÓN: morinda citrifolia L (Noni) ha ganado popularidad entre la población cubana, lo que sugiere la necesidad de caracterizar farmacológicamente las preparaciones de noni desarrolladas en Cuba. OBJETIVO: evaluar los efectos de extractos de hojas y frutos de Noni sobre la agregación plaquetaria y la coagulación sanguínea, así como la influencia de la edad de la planta, la madurez de la hoja y los contenidos de fenoles totales y flavonoides. MÉTODOS: hojas verdes y amarillas y frutos ligeros amarillos fueron recolectados de plantas de noni de tres y seis años de edad. Extractos de hojas (30 g/100 mL w/v) y frutos (100 g/100 mL w/v) estuvieron preparados por maceración en alcohol al 50 % y agua, al respecto. Las concentraciones de sólidos totales (TSS), compuestos fenólicos (PHEN) y flavonoides expresados como quercetina (FLV) fueron determinadas. Los efectos de los extractos de hojas (0,4 y 0,8 mg TSS/mL) y de frutos (0,6 y 1,2 mg TSS/mL) sobre la agregación plaquetaria (PA) inducida por ADP, el tiempo de protrombina (PT) y el tiempo parcial de tromboplastina parcial activado (PATT) fueron determinados en plasma humano in vitro y los efectos de un extracto de hojas (42 y 378 mg TSS/kg i.p. y 630 mg/kg p.o.) sobre estas variables de plasmas de ratas fueron evaluados ex vivo. RESULTADOS: los extractos de frutos tuvieron mayor contenido de TSS pero menores de PHEN y FLV que los de hojas. Estos últimos inhibieron la PA in vitro en plasma humano independiente de la edad de la planta y la madurez de las hojas. Un extracto de hojas indujo significativa reducción de PA ex vivo en ratas. TP y PATT no fueron modificados en ninguna situación experimental. CONCLUSIÓN: extractos hidroalcohólicos de hojas de Noni desarrollados en Cuba tienen efecto antiagregante plaquetario pero no anticoagulante.


INTRODUCTION: morinda citrifolia L (Noni) is a medicinal plant species that has gained popularity among Cuban population, suggesting the need to characterize the pharmacological activities of noni preparations developed in Cuba. OBJECTIVE: assessing the effects of Noni leaf and fruit extracts on platelet aggregation and blood coagulation, as well as the possible influence of plant age, leaf maturity and extract total phenol and flavonoid contents on it. METHODS: green and yellow leaves and light yellow fruits were collected from three- and six- year old Noni trees. Leaf (30 g/100 mL w/v) and fruit (100 g/100 mL w/v) extracts were prepared by maceration in 50 % alcohol and water respectively. The concentrations of total soluble solids (TSS), phenolic compounds (PHEN) and flavonoids expressed as quercetin (FLV) in the extracts were determined. The in vitro effects of leaf (0,4 and 0,8 mg TSS/mL) and fruit (0,6 and 1,2 mg TSS/mL) extracts on ADP-induced platelet aggregation (PA), prothrombin time (PT) and partial activated thromboplastine time (PATT) in human plasma was determined. Furthermore, the ex vivo effects of a leaf extract (42 and 378 mg TSS / kg i.p. and 630 mg/kg p.o.) on these variables of rat plasma were assessed. RESULTS: fruit extracts showed higher TSS, but lower PHE and FLV values than leaf extracts. Leaf but not fruit extracts inhibited ADP-induced PA that was independent on plant age and leaf maturity. A significant reduction of PA was ex vivo induced by a leaf extract in rats. TP and PATT were not modified during in vivo or ex vivoexperiments. CONCLUSION: noni leaf hydro-alcohol extracts developed in Cuba have antiplatelet but not anticoagulant effect.

ABSTRACT

O objetivo deste trabalho, relacionado a produção orgânica, utilizou preparados homeopáticos visando incremento na biomassa e no teor de flavonóides em plantas dióicas de carqueja. Os tratamentos foram: controle com água, controle com álcool 70%, Phosphorus 12CH, Sulphur 6CH, e a combinação destas homeopatias. Variações fenotípicas foram registradas a partir da análise de crescimento, realizada quinzenalmente durante 60 dias e dos caracteres morfológicos, tais como: como altura, número de ramificações, comprimento do ramo principal, diâmetro do caule e ala caulinar mediana. Na colheita foram medidos: a fitomassa fresca/seca e o volume de raiz. Posteriormente, foi quantificado o teor de flavonóides totais conforme as análises prescritas pela Farmacopéia Brasileira. O experimento foi avaliado em delineamento de blocos casualizados (2x5x2), sendo suas médias avaliadas pelo teste Tukey e o coeficiente de variação ambiental para estimar a plasticidade. Quanto ao estudo do crescimento da planta ao longo do tempo, verificou-se aumento linear simples para a maioria das variáveis nos dois sexos. Foram detectadas respostas plásticas na maioria dos caracteres avaliados. De acordo com os resultados, em ambos os sexos, 60 dias de cultivo são suficientes para obtenção de matéria prima com teor aceitável de flavonóides, bem como para verificação dos efeitos de patogenesia e similitude na aplicação de preparados homeopáticos Phosphorus 12CH, Sulphur 6CH e sua combinação Phosphorus 12CH + Sulphur 6CH. A fitomassa e o teor de flavonóides totais foram semelhantes entre plantas dióicas durante o período de estudo.


This study looked at organic production, in addition to the use of homeopathic preparations, in order to increase biomass and flavonoid content in dioecious plants of Baccharis genistelloides. The adopted treatments were: water control, control with 70% alcohol, Phosphorus 12CH, Sulphur 6CH and a combination of them. Phenotypic changes were recorded for the growth analysis, performed biweekly for 60 days, of the morphological features, such as height, number of branches, length of the main branch, diameter of the stem and median stem wing. At harvest, we measured the fresh/dry phytomass and root volume. Subsequently, we quantified the content of total flavonoids according to the analyses required by the Brazilian Pharmacopoeia. The experiment was evaluated in a randomized block design, and its averages were evaluated by the Tukey's test and environmental variation coefficient to estimate plasticity. As for the study of plant growth over time, there was a simple linear increase for most of the variables in both genders. We could detect phenotypic plasticity traits in most features analyzed. According to these results in both genders, 60 days after inoculation are sufficient to obtain raw material with acceptable level of flavonoids, as well as pathogenesis and similar effects in the application of homeopathic preparations of Phosphorus 12CH, Sulphur 6CH and their combination. Phytomass and content of total flavonoids were similar between the dioecious plants during the study period.

ABSTRACT

Cerebrovascular accidents and coronary artery disease are the leading causes of cardiovascular mortalities in Brazil and high levels of LDL cholesterol are one of the main risk factors. In this context, several plant extracts and natural substances have shown promise as cholesterol-lowering. The objective of this study was to evaluate the potential of the hydroalcoholic extract of the fruit of H. dulcis and of dihydromyricetin in cholesterol reduction in hypercholesterolemic rats. Forty-two Wistar male rats were distributed into seven groups of six animals that received diets supplemented with 1% cholesterol and 0.3% cholic acid, with the exception of the control group, which received conventional diets. Animals were treated with oral suspensions containing: atorvastatin 1.0 mg/kg; H. dulcis extract at 50.0 and 100.0 mg/kg and dihydromyricetin at 25.0 and 50.0 mg/kg vehicle (control group). The following biochemical markers were evaluated; total cholesterol, HDL-C, LDL-C, triglycerides, AST, ALT, and alkaline phosphatase. The hypercholesterolemic diet was effective in inducing hypercholesterolemia, increasing total cholesterol by 112.7% relative to the control group. The treatments with two doses of the extract proved to be promising hypocholesterolemic agents, as they were able to substantially reduce total cholesterol and LDL-C, without significantly altering triglycerides, hepatic transaminases, and alkaline phosphatase, thereby encouraging the studies with the plant H. dulcis. The groups treated with the flavonoid dihydromyricetin, although they showed a significant reduction in total cholesterol and LDL-C, and found increases in triglycerides and hepatic transaminases, which is unwanted in the context of hypercholesterolaemia.


No Brasil, o acidente vascular cerebral e a doença arterial coronariana constituem as principais causas de mortalidade cardiovascular, sendo os altos níveis de colesterol LDL um dos principais fatores de risco. Nesse contexto, diversos extratos vegetais e substâncias naturais isoladas têm se mostrado promissoras como hipocolesterolemiantes. O objetivo do trabalho foi avaliar o potencial do extrato hidroalcoólico dos frutos de Hovenia dulcis e do flavonóide diidromiricetina na redução do colesterol em ratos hipercolesterolêmicos. Quarenta e dois ratos Wistar machos, foram distribuídos em 7 grupos de 6 animais, que receberam dieta suplementada com 1% de colesterol e 0,3% de ácido cólico, à exceção do grupo controle, que recebeu ração convencional. Posteriormente, os animais foram tratados com suspensões orais contendo: atorvastatina 1,0 mg/kg; extrato de H. dulcis de 50,0 e 100,0 mg/kg; diidromiricetina de 25,0 e 50,0 mg/kg e veículo (grupo controle). Avaliaram-se os parâmetros bioquímicos: colesterol total, HDL-C, LDL-C, triglicérides, AST, ALT e fosfatase alcalina. A dieta hipercolesterolêmica foi efetiva na indução da hipercolesterolemia, aumentando o colesterol total em 112,7% em relação ao controle. Os tratamentos com as duas doses do extrato mostraram-se promissores como agentes hipocolesterolemiantes, já que foram capazes de reduzir substancialmente o colesterol total e LDL-C, sem alterar significativamente triglicérides, as transaminases hepáticas e a fosfatase alcalina, incentivando, assim, a continuidade de estudos com a planta H. dulcis. Já os grupos tratados com o flavonóide diidromiricetina, apesar de apresentarem redução significativa do colesterol total e de LDL-C, apresentaram elevações nos triglicérides e nos parâmetros hepáticos, resultado indesejável no âmbito das hipercolesterolemias.

ABSTRACT

The flavonoids are a large class of polyphenolic compounds found in plants that are known to exhibit biological effects. In the study, the flavonoid 5.7.4’-trimethoxyflavone (TMF) extracted from Praxelis clematidea was evaluated for its antibacterial activity. Microdilution method was used for antibacterial assay of the flavonoid and eleven bacteria strains were used in the study for activities. The results were also compared with the standard drug, Chloramphenicol (100 ug/mL). The results obtained showed activity of the flavonoid against Gram positive and Gram negative bacteria.


Los flavonoides son una clase importante de compuestos polifenólicos encontrados en las plantas que se sabe que presentan efectos biológicos. En el estudio, el flavonoide 5.7.4 '-trimethoxyflavone (TMF) extraído de Praxelis clematidea fue evaluado por su actividad antibacteriana. Se utilizó el método de microdilución para el ensayo antibacteriano del flavonoide y once cepas de bacterias se usaron en el estudio de las actividades. Los resultados se compararon también con el fármaco estándar, Cloranfenicol (100 ug/mL). Los resultados obtenidos mostraron actividad del flavonoide contra bacterias Gram positivas y Gram negativas.

ABSTRACT

Introducción: Las dislipidemias representan un factor de riesgo primario para la cardiopatía coronaria. Objetivo: Comparar el efecto sobre el perfil lipídico entre Averrhoa carambola L. o "carambola" vs el Gemfibrozilo en Rattus rattus var albinus. Material y Métodos: Se realizó un estudio aleatorizado. Se trabajó con 39 Rattus rattus var albinus machos; divididos al azar en 2 grupos experimentales (GE) y un grupo control (GC). Sometidos a 2 semanas de acondicionamiento, 2 semanas de alimentación rica en grasa (PIG); posteriormente se administró carambola y Gemfibrozilo a grupos experimentales (GE1 y GE2, respectivamente). Se midió el perfil lipídico en cada etapa y se aplicó la Prueba de Análisis de Varianza y prueba de Duncan. Resultados: El nivel de colesterol sérico: GC basal 80 mg/dL, PIG 97 mg/dL y a las dos semanas 83 mg/dL. En GE1 basal 84 mg/dL, PIG 100 mg/dL, y a las dos semanas 72 mg/dL (p<0.05). En GE2 basal 83 mg/dL, PIG 104 mg/dL y a las dos semanas 80 mg/dL. El nivel de LDL Colesterol sérico: GC basal 47 mg/dL, PIG 66 mg/dL y a las dos semanas 50 mg/dL. GE1 basal 46 mg/dL, PIG 65 mg/dL, y a las dos semanas 39 mg/dL(p<0.05). GE2 basal de 47 mg/dL, PIG de 71 mg/dL y a las dos semanas 42 mg/dL. Los triglicéridos y el HDL no tuvieron cambios significativos por la carambola pero sí por el Gemfibrozilo. Conclusiones: La ingesta de carambola disminuye significativamente los niveles de colesterol total y colesterol LDL.


Introducción: Dyslipidemia represent a primary risk factor for coronary heart disease. Objectives: Compare the effect on the lipid profile between Averrhoa Starfruit, "Starfruit" versus Gemfibrozile in Rattus rattus var albinus. Material and Methods: A randomized study was realized. I worked with 39 Rattus rattus var albinus male; were divided in 2 experimental groups and a group control (GC). They were put under stage of preparation by 2 weeks, alter that, they were feeding with a rich fat diet (PIG) for 2 weeks; later I administered Starfruit and Gemfibrozile to experimental groups (GE1 and GE2, respectively). The lipid profile in each stage was measured and it was applied to the Test of Variance Analysis and test of Duncan. Results: The seric cholesterol level: GC basal 80mg/dL, PIG 97mg/dL, at two week 83 mg/dL. GE1 basal 84 mg/dL, PIG 100 mg/dL, at two week 72 mg/dL(p<0.05). GE2 basal 83 mg/dL, PIG 104mg/dL, at two weeks 80 mg/dL. The seric level of LDL Cholesterol: GC basal 47 mg/dL, PIG 66 mg/dL, at two weeks 50mg/dL. GE1 basal 46mg/dL, PIG 65 mg/dL, at two weeks 39 mg/dL (p<0.05). GE2 basal 47 mg/dL, PIG 71 mg/dL, at two weeks 42 mg/dL. The seric level of Triglycerides and The seric level of HDL Cholesterol was not significantly changes for de starfruit but by gemfibrozile. Conclusions: The Starfruit ingestion significantly diminishes the levels of total cholesterol, cholesterol LDL in Rattus rattus var albinus.

ABSTRACT

La fracción butanólica del extracto de Cecropia peltata L, (N. V.: "Yarumo"), especie medicinal utilizada por la tradición popular en Colombia con fines tranquilizantes, presenta en ratones albinos ICR un perfil de tipo anticonvulsivante, ansiolítico y sedante, según las pruebas de electroshock, pentilentetrazol, ocultamiento de esferas y potenciación de sueño barbitúrico (80 mg/kg, v. o., p = 0,05 vs. control). Paralelamente, carece de efectos de tipo locomotor y antidepresivo, según las pruebas del alambre y nado forzado. El efecto de esta fracción fue superior al presentado por el extracto etanólico y las fracciones acuosa y hexánica. Además, la subfracción butanólica rica en flavonoides, presenta un efecto "dosis-respuesta" en la prueba de convulsión máxima inducida por electroshock (80 mg/kg - 240 mg/kg). Estos hallazgos sugieren que los metabolitos de tipo flavonoide podrían ser responsables de la actividad farmacológica de Cecropia peltata y dan soporte al uso tradicional de esta especie.


The butanolic fraction obtained from the extract of Cecropia peltata L (NV: "Yarumo"), medicinal plant used by popular tradition in Colombia as tranquilizer, displays in ICR albino mice an anticonvulsant, anxiolytic and sedative profile, according to electroshock, pentylenetetrazole, marble burying and barbiturate sleeping tests (80 mg/kg, p. o., p = 0.05 vs. control). At the same time Cecropia peltata does not to seem to alter the locomotor function nor to have antidepressant effect, according to their effect in wiring and forced swimming tests. The effect of this fraction was greater than ethanolic, aqueous and hexanic fractions. Additionally, a sub-fraction from butanolic part, rich in flavonoids, elicits a "dose-response" effect in maximum electroshock-induced seizures (80 - 240 mg/kg). These findings suggest that flavonoid metabolites play a key role in the pharmacological activity of Cecropia peltata and support the traditional use of this species.

ABSTRACT

The antioxidant activity of resinous extracts obtained from H. stenophylum and H. sinuatum species, was evaluated through ORAC index (Oxygen Radical Absorbance Capacity) in water phase and in presence of Triton X-100 micelles, using as test molecules to pyrogallol red (PGR) and evaluating their reduction by the action of peroxyl radicals obtained from thermolysis of AAPH. The results show that these extracts protect to PGR of the action of the radicals. This protection is reduced drastically in the presence of Triton X-100 micelles. The same effect was observed with the main flavonoid of these extracts (3-O-methylgalangin). These results show the importance of the media of reaction of pure compounds and/or extracts at the time of to take into account their use as antioxidants.


La actividad antioxidante de exudados resinosos obtenidos desde las especies H. stenophylum y H. sinuatum, fue evaluada a través del ensayo ORAC (Oxygen Radical Absorbance Capacity) en fase acuosa y en presencia de micelas de Triton X-100, usando como molécula prueba a pirogalol rojo (PGR) y evaluando su reducción frente a la acción de radicales peróxidos obtenidos desde la termólisis de AAPH. Los resultados muestran que estos extractos protegen al PGR de la acción de los radicales. Esta protección es reducida drásticamente en presencia de micelas de Tritón X-100. El mismo efecto fue observado con el flavonoide mayoritario de estos extractos (3-O-metilgalangina). Estos resultados muestran la importancia de considerar el medio de reacción de compuestos puros y/o extractos al momento de tomar en cuenta su uso como antioxidantes.

El própolis es una resina cérea de composición compleja y consistencia viscosa, que usado en la alimentación de animales, favorece el incremento de peso y masa muscular, mediante la estimulación de la ingesta alimenticia. En cabras estimula la producción de sólidos totales, grasa y proteína en la leche; resultando benéfico también en bovinos, pollos, porcinos, tilapias y trucha arco iris. El propóleo al ser evaluado en diferentes especies animales, ha demostrado poseer propiedades inmunoestimulantes, antioxidantes, antibacteriales, antitumorales y antivirales. Estas características hacen del propóleo una sustancia importante, que puede ser incluida en la dieta de los animales para fortalecer su sistema inmunológico, por su resistencia a enfermedades y ayudan a mejorar el rendimiento productivo, características que lo clasifican como aditivo natural funcional.


Propolis is a waxy resin of a complex composition and a viscose consistency, that used in animal feeding, favors weight increase and muscle mass gain, by stimulation of food intake. In goats it stimulates production of total solid, fat and milk proteins. It also shows beneficial results in bovines, broilers, pigs, tilapia and rainbow trout. Propolis, when evaluated in different animal species, has shown to have immunostimulatory, antioxidant, antibacterial, antiviral and antitumor properties. These characteristics make of propolis an important substance that can be included in the animal diet in order to strengthen their immune system, because of its resistance to disease and because they help improve production efficiency, characteristics that classify it as a functional natural additive.

O fracionamento do extrato metanólico das folhas de Gochnatia barrosii Cabrera (Asteraceae) resultou no isolamento do flavonol glicosídico trans-tilirosídeo [kaempferol 3-O- -D-(6''-O-E-p-cumaroil)-glicopiranosídeo], que nunca tinha sido identificado na referida espécie vegetal. Em teste realizado in vitro, observou-se que tal substância a 500 μg/mL, não tem efeito sobre a mortalidade de juvenis do segundo estágio do nematóide Meloidogyne exigua Goeldi.


Fractionation of the methanolic extract from Gochnatia barrosii Cabrera (Asteraceae) leaves resulted in the isolation of the flavonol glycoside trans-tiliroside [kaempferol 3-O- -D-(6''-O-E-p-coumaroyl)-glucopyranoside], which had never been found in such plant species. Such substance at 500 μg/mL caused no in vitro effect on the mortality of second-stage juveniles of the nematode Meloidogyne exigua Goeldi.

El propóleo, es una sustancia compleja, de origen vegetal, que preparan las abejas a partir de la recolección de resinas producidas en algunas plantas (principalmente árboles). Una de las actividades más importantes del propóleo es su actividad antimicrobiana la cual se le atribuye básicamente a los flavonoides, es un compuesto bioactivo de grandes potencialidades para el tratamiento como antiséptico de aftas en la boca, herpes, amigdalitis, ayuda en la cicatrización de las heridas, antiinflamatorio, anticaries, cirugía oral, endodoncia, periodoncia y patología oral entre otras. Es importante destacar que existen diversos estudios enfocados en la eliminación de la caries dental relacionados con el propóleo con el fin de determinar la inhibición del crecimiento de varios microorganismos bucales, los cuales han sugerido que esta sustancia natural interfiere en la inhibición de la actividad enzimática de varias proteínas que so necesarias para el crecimiento y desarrollo de los mismos, contribuyendo a la posible solución de un problema de salud pública como es la caries dental. Esta revisión está basada en proporcionar una visión actualizada sobre el uso del propóleo en el área de odontología


The propolis is a complex substance of plant origin; is a resinous mixture that honey bees collect from tree buds and other plants, (primarily trees). Its antimicrobial activity is one of the most important ones, which is mainly attributed to flavonoids. Propolis is an effective bioactive compound used as an antiseptic for the treatment of aphte in the mouth, herpes and amygdalitis. It is also used for injury healing, as an anti- inflammatory, anticaries, oral surgery, endodontic, periodontics and oral pathology, among others. It is worth noting, there are several studies on the elimination of cavities relating to propolis in order to determine its effect as a growth inhibitor on numerous oral microorganisms. These studies make us believe that this natural substance interferes with the inhibition of the enzymatic activity of several proteins that are necessary for their growth and development of microorganisms, contributing as a solution for dental cavities which is a public health problem. This review is aimed at providing an updated viewpoint on the use of propolis in dentistry

ABSTRACT

This paper presents the chemical investigation of the leaves and stems of Ouratea castaneifolia (DC.) Engl.. There are no chemical or pharmacological studies with this species. Classic phytochemical investigation of the organic extracts together with high pressure liquid chromatography (HPLC) procedures lead to the identification of seventeen metabolites: seven triterpenes (friedelin, 3β-friedelinol, α-amyrin, β-amyrin, lupeol, germanicol and taraxerol), four steroids (sitosterol, stigmasterol and the glycosides sitosteryl 3-O-β-D-glucopyranoside and stigmasteryl 3-O-β-D-glucopyranoside), one isoflavone (5,7,4'-trimethoxyisoflavone), one flavone (5,4'-dihydroxy-7,3',5'-trimethoxyflavone) and four biflavones (amenthoflavone, 7,7"-O-dimethylamenthoflavone, heveaflavone and tetramethylamenthoflavone). The structures of the compounds were established by the analysis of ¹H, 13C NMR spectra including bidimensional techniques. The classes of the identified metabolites are in agreement with previous studies of the Ouratea genus.


O presente trabalho trata da investigação química das folhas e caule da espécie Ouratea castaneifolia (DC.) Engl., sobre a qual não há registros de estudos químicos ou farmacológicos anteriores. O estudo fitoquímico clássico dos extratos orgânicos do caule e das folhas de O. castaneifolia foi aliado à técnica da cromatografia líquida de alta eficiência (CLAE) e resultou na identificação de dezessete metabólitos: sete triperpenos (friedelina, 3β-friedelinol, α-amirina, β-amirina, lupeol, taraxerol e germanicol), quatro esteróides (sitosterol, estigmasterol e os glucosídeos sitosteril 3-O-β-D-glicopiranosídeo e estigmasteril 3-O-β-D-glicopiranosídeo), uma isoflavona (5,7,4´-trimetoxiisoflavona), uma flavona (5,4´-diidroxi-7,3´,5´-trimetoxiflavona), quatro biflavonas (amentoflavona, 7,7"-O-dimetil-amentoflavona, heveaflavona e tetrametilamentoflavona). A identificação das substâncias foi feita com base na análise de espectros de RMN de ¹H, 13C e técnicas bidimensionais. As classes dos metabólitos identificados estão de acordo com aquelas citadas em estudos químicos do gênero Ouratea.

ABSTRACT

Grammosciadium platycarpum Boiss. & Hausskn (Apiaceae) is one of three endemic Iranian species of the genus Grammosciadium DC. Consumption of the aerial parts of this plant affects renal function and causes diuresis. In the DPPH assay the methanol extract showed the highest level of free radical scavenging activity (RC50 = 1.196 x 10-2 mg/mL) among the extracts. Reversed-phase preparative HPLC analyses of the methanol extract yielded nine flavonoids, which were responsible for the free radical scavenging activity of the MeOH extract. The GC-MS analyses of the essential oils led to the identification of 29 terpenoids, mainly monoterpenes (non-oxygenated 3.97 percent and oxygenated 77.49 percent) accounting for over 96 percent of the total oils.


Grammosciadium platycarpum Boiss. & Hausskn (Apiaceae) é uma das três espécies endémicas no Irã do gênero Grammosciadium DC. Consumo da parte aérea da planta afeta a função renal e causa diurese. No ensaio por DPPH o extrato metanólico apresentou o maior nível de atividade de antioxidante por radicais livres (RC50 = 1,196 x 10-2 mg/mL) entre os extratos. Análises por HPLC preparativa de fase reversa do extrato metanólico resultou no isolamento de nove flavonóides, os quais foram responsáveis pela atividade de antioxidante do extrato metanólico. A análises por CG-EM dos óleos essenciais levou à identificação de 29 terpenóides, principalmente monoterpenos (não-oxigenados 3,97 por cento e oxigenados, 77,49 por cento), os quais representam mais de 96 por cento do total de óleos.

ABSTRACT

Os Flavonóides são substâncias naturais amplamente distribuídas no Reino Vegetal. Há um interesse crescente na investigação dessas substâncias, devido ao volume de evidência dos benefícios que eles proporcionam para a saúde. A Rutina é um flavonol glicosídico pertencente a uma importante classe de Flavonóides, sendo extensamente encontrados na natureza, em frutas, vegetais e bebidas como chá e vinho. Pouco se sabe sobre a farmacocinética e biodisponibilidade da rutina, e estes mecanismos tem sido foco de muita controvérsia. Apesar disso, a Rutina apresenta grande importância terapêutica por melhorar a resistência e permeabilidade dos vasos capilares, atividades antioxidante, antiinflamatória, anticarcinogênica dentre outras, razão que levou à realização da presente revisão.


Flavonoids are natural products widely distributed in the vegetable kingdom. There has been increasing interest in research of flavonoids, due to growing evidence of the health benefits of them. Rutin is a glycoside flavonol which belongs to an important class of flavonoids, being extensively found in the nature in fruit, vegetable and beverages, such as tea and wine. About the pharmacokinetics and bioavailability of Rutin, little is stil know, and tis mechanisms have been a matter of much controversy. Dispite it, Rutin presents a therapeutical importance due to improve resistance and permeability of capillaries vessels, antioxidante, antiinflamatory, anticarcinogenic and other important activities. Reason that led to the completion of this review.

ABSTRACT

Conocliniopsis prasiifolia (DC) R.M. King & H. Robinson é uma espécie utilizada na medicina popular como antiinflamatória e analgésica, pertencente à família Asteraceae e conhecida como "mentrasto-roxo". Estudos prévios com o extrato etanólico das folhas de C. prasiifolia (EECp) demonstraram uma baixa toxicidade por via oral e intraperitoneal, bem como efeito analgésico e antiinflamatório em modelos animais. O presente estudo objetivou avaliar o efeito de EECp frente à degranulação de neutrófilos, e dessa forma melhor caracterizar o efeito antiinflamatório dessa espécie vegetal. Para tanto, foram utilizados neutrófilos peritoneais de ratos e técnicas espectrofotométricas. Os resultados demonstraram que EECp, nas concentrações de 25 e 50 µg/mL, foi capaz de inibir de forma significativa (p<0,05) a degranulação de neutrófilos induzida por fMLP. Portanto, o EECp impede a liberação de grânulos dos neutrófilos contendo enzimas degradativas, demonstrando atividade antiinflamatória.

Studies of the ethanol extract from its leaves (EECp) showed a low toxicity throughout oral and intraperitoneal routes, and anti-inflammatory and analgesic effects in animal models. The aim of this study was to evaluate the effect of EECp in the neutrophils degranulation for better characterization of its anti-inflammatory effect. Peritoneal neutrophils from rats and spectrophotometric techniques were used. The results showed that the EECp (25 and 50 µg/mL) was able to inhibit significantly (p<0.05) the neutrophils degranulation induced from fMLP. We can conclude that the EECp prevents the release of granules of the neutrophils that contains degrading enzymes, demonstrating anti-inflammatory activity.
